Orforglipron (developed by Eli Lilly) is a first-in-class oral non-peptide GLP-1 receptor agonist — meaning it is a small molecule that activates GLP-1 receptors without being a peptide itself. Unlike oral semaglutide (Rybelsus), which requires strict fasting conditions and has limited bioavailability, orforglipron can be taken at any time without food restrictions. In Phase 2 GZLD trials, orforglipron produced approximately 14.7% body weight reduction over 36 weeks in adults with obesity — comparable to injectable semaglutide. It also demonstrated significant HbA1c reductions in patients with type 2 diabetes. As a fully oral agent with no injection burden, orforglipron has the potential to dramatically expand access to GLP-1 class therapy. Phase 3 trials are underway.
